Risk Factors and Mechanisms of Alzheimer's Disease Pathogenesis: Obviously and Obviously Not.

چکیده
In their article, Chen and Fernandez (3) present a viewpoint in which they briefly consider the roles of the aging process, and risk factors associated with age-related degenerative diseases, in the pathogenesis of Alzheimers disease (AD). The authors forward both the general perspective, and a specific mechanism which they suggest underlies most cases of AD. They propose that 1) several known risk factors most likely play a critical role in the late-onset sporadic AD, and 2) plaques and tangles occur spontaneously during aging as a result of a natural decline of energy metabolism and Ca signaling. My responses to these two major conclusions are that 1) obviously, genetic and environmental risk factors play important roles in the pathogenesis of AD, and this is certainly not a new perspective; and 2) the proposal that a decline in energy metabolism and Ca signaling is central to the pathogenesis of AD is obviously not correct. In the following paragraphs I will elaborate on these two points. It is obviously the case that there are several important risk factors that increase the probability of developing AD. The aging process is the strongest risk factor for AD, and the molecular and cellular changes that occur in the brain dur-

Alzheimer's desease is the most common age-related neurodegenerative disorder, and cognitive problems such as defects in learning and memory are of its symptoms. Among the factors involved in the pathogenesis of the disease are biochemical disorders in protein production, oxidative stress, decreased acetylcholine secretion and inflammation of the brain tissue. Extra-neuronal accumulation ...
متن کاملRisk Prediction of Leptospirosis by Considering Environmental Factors in Iran Using MAXENT Model
The global burden of leptospirosis as a fatal zoonotic disease is increasing all over the world [1]. As there is not any significant decrease in yearly reported cases trend in Iran and potential spatial distribution of leptospirosis remain unknown in national level, we tried to figure out the geographic distribution pattern of leptospirosis in all parts of Iran. The aim of this study is produci...

For a long period of time some different hypothesis have been published about arteriosclerosis but none of them has discovered the pathogenesis and effective treatment of this entity. By elemination of risk factors it is not clear that the disease process stops or reverses. Preventiona and treatment of risk factors should start before manifestation of arteriosclerosis sympthoms. The result of...

Osteoarthritis (OA) a common disease of aged population and one of the leading causes of disability. Incidence of knee OA is rising by increasing average age of general population. Age, weight, trauma to joint due to repetiting movements in particular squatting and kneeling are common risk factors of knee OA. Several factors including cytokines, leptin, and mechanical forces are pathogenic fact...
متن کاملVitamin D level in Alzheimer\'s disease: a case control study
Background: Alzheimer's disease (AD) is a chronic neurodegenerative disease characterized by a progressive decline of cognitive performance, which has a harmful impact on social activities. AD is the main cause of dementia and loss of functional independence in the elderly. AD is a worldwide concern because of its adverse consequences and expanding prevalence and incidence. Vitamin D is the mos...
